چکیده

Putrefactive Anaerobe strain 3679 (PA 3679) is of interest to the food microbiologist because it has been responsible for the microbial spoilage of certain types of thermally processed foods. It is also of interest because the spores of this organism have a high degree of heat resistance and their thermal death rate is frequently used in the calculation of thermal processes for low acid canned foods. Previous procedures for producing spores of PA 3679 have required complex media and long incubation periods: Reed et al. (1951), Heiligman et al. (1956), McClung (1935), National Canners Association (1954), and Roth and Halvorson (1952). In addition, many of these media contain particulate material which is difficult to remove from the spore harvest. The objectives of this investigation were two-fold: (a) to develop a clear medium which would stimulate vegetative growth as well as maximum sporulation within shorter incubation periods and (b) to devise a cleaning procedure, without the use of heat, which would effectively remove the vegetative cells and debris from the harvested spore crop without impairing the viability of the treated spores.

The influence of recovery media on thermal resistance values of spores of a Putrefactive Anaerobic Bacterium.

Yesair's pork infusion medium, as modified by Wynne and Foster (1948), has been extensively employed in thermal resistance studies of spores of Clostridium species. While this medium is excellent for the recovery of spores of these organisms, it is expensive and laborious to prepare. Recent unpublished data in this laboratory have demonstrated that counts of heat-activated spores of Putrefactiv...

The influence of various media on spore count determinations of a putrefactive anaerobe.

Putrefactive anaerobe no. 3679 has been widely used as a test organism in thermal resistance studies. The choice of media for spore production, spore count determinations, and thermal resbtance studies has been varied by practically all workers employing this organism. Since an accurate estimation of thermal resistance is dependent upon a knowledge of the number of spores initially present, it ...

Method for Obtaining Cleaned Putrefactive Anaerobe 3679 Spores.

Sporulating clostridial suspensions tend to recycle through subsequent phases such as germination, outgrowth, and further multiplication. The result is a mixture of vegetative cells and of spores at various developmental stages. Recently, activated-culture techniques have been used to achieve almost complete sporulation in semisynchronized populations of several clostridia (Collier, 1). 10, in ...
